The aim of the project is to create new mucoadhesive polymeric gel and gel-forming in situ smart materials of prolonged proteolytic, fibrinolytic, collagenolytic, anti-inflammatory action with immobilized serratiopeptidase and antiseptic drugs, research of their biochemical and physicochemical properties, implementation of medical and biological testing. The objectives of the project. 1) Develop bioadhesive composite matrices using polymers of natural and synthetic origin: carbopol, alginates, cellulose derivatives to include the enzyme serratiopeptidase with complex proteolytic activity and antiseptic agents. Investigate the conditions of gelation in situ due to the ion- or thermoinitiated method of obtaining a gel. 2) Immobilize the enzyme and antiseptic agents in bioadhesive matrices with a detailed study of the interaction of BAR-polymer using physicochemical methods and the influence of pH, temperature, ratio of reagents. Evaluate the effectiveness of the inclusion of serratiopeptidase according to the determination of protein content and preservation of proteolytic, fibrinolytic and collagenolytic activities. 3) To establish physicochemical and biochemical features of serratiopeptidase immobilized into bioadhesive polymer gels and gel-forming smart materials: sorbent capacity, mucoadhesion to mucous membrane, pH and thermal dependence of activity, stability and other characteristics of bioactive polymers. Choose the most promising preparation for further research. 4) Conduct medical and biological tests of the developed bioadhesive polymeric material: microbiological researches, anti-inflammatory and antimicrobic action. Prepare scientific and technical documentation (draft technical conditions) and report. The work will be aimed at solving fundamental problems and practical issues of bioorganic and medical chemistry, engineering enzymology, biochemistry, biotechnology, pharmacology to obtain modern biomaterials promising for use in medicine.
